Maintenance of genome integrity during DNA replication

Leader: Philippe PASERO


A central question in cancer biology is how genomic instability arises in normal cells. Recent evidence indicates that replication stress is detected very early during the cancer process and contributes to the development of the disease. Our research focuses on the origin of this replication stress in yeast and in human cells. We use powerful new technologies such as DNA combing and ChIP-on-chip to monitor replication fork arrest and recovery at the level of individual molecules and on a genome-wide basis. These studies should provide new insights into how DNA replication errors promote the development of cancer and of other genetic disorders associated with increased genomic instability.
